In my little clinic, we do the PCR in three main steps on our old machine. First, denaturation at 95°C to split the DNA, then annealing at around128 55°C for primers to stick, and finally extension at 72°C to build the new strand. But sahib, with our budget machine, the annealing temperature is never so precise—it often needs adjusting by trial and error, like finding the right flame for my morning chai. This I know from squinting at faded numbers while my son explains the manual over a crackling phone call from Lucknow.
